G-CSF is a hematopoietic growth factor that has
been used to mobilize stem/progenitor cells into the
blood of donors for allogeneic transplantation in
patients with malignant hematologic and lymphoid
disorders . G-CSF also mobilizes human dendritic
cells with the capacity to polarize na€ ı ıve CD4 T
cells toward a Th2 phenotype . Recently, G-CSF
treatment was able to provide protection of mice with
ongoing experimental autoimmune encephalomyelitis,
via chemokine and cytokine based immune deviation
. G-CSF treatment of mice polarizes T cells toward
a Th2 pattern, and mobilizes APC expressing MHC-II
molecules with low levels of costimulatory molecules
. Interactions between T cells and APC without
appropriate costimulation can lead to anergy and tol-
erance . In addition, monocytes from G-CSF
treated mice can suppress T cell proliferation via IL-10
production . Dendritic cells from G-CSF treated
mice and humans had decreased production of TNF-a
or IL-12 after in vitro activation .
These findings prompted us to investigate whether
immune polarization and/or tolerogenic properties of
G-CSF mobilization might be of therapeutic benefit in
the treatment of adjuvant arthritis , a model of
autoimmune arthritis that has some features in common
with inflammatory arthritis in humans . AA can
be induced in Lewis rats by the subcutaneous injec-
tion of a suspension of Mycobacterium tuberculosis in
mineral oil. In this model, the reactivity of T cells to
mycobacterial antigens has been implicated in the
pathogenesis of joint inflammation . In fact, AA
can be induced in Lewis rats with a single T cell clone
that recognizes the peptide 180–188 of the 65 kDa my-
cobacteria heat-shock protein (hsp65) and has been
shown to cross-react with a fraction of cartilage pro-
teoglycan .
The critical role of the 180–188 sequence in AA in the
Lewis rat, became more apparent when it was shown
that adjuvant arthritis can be prevented by administer-
ing peptide 180–188 subcutaneously in mineral oil
or peptide 176–190 intranasally . Therefore,
we evaluated the effects of G-CSF mobilization in
addition to peptide administration in the treatment of
AA in Lewis rats.
